A glia cortical em camundongos SOD1 (G93A) é sutilmente afetada pela ELA

O papel da glia na esclerose lateral amiotrófica (ELA) é inegável. Sua atividade relacionada à doença foi extensivamente estudada na medula espinhal, mas apenas parcialmente no cérebro. Apresentamos aqui um estudo abrangente da glia no córtex de camundongos SOD1 (G93A) - um modelo amplamente utilizado de ALS. Utilizando sequenciamento de RNA unicelular (scRNA-seq) e imuno-histoquímica, inspecionamos astrócitos, micróglia e oligodendrócitos, em quatro estágios da doença, respeitando o fator sexo. Relatamos alterações mínimas da glia ao longo da progressão da doença e independentemente do sexo. As análises de pseudovolume e de células únicas revelaram alterações transcricionais sutis relacionadas à doença no estágio final na micróglia e nos oligodendrócitos, que foram corroboradas pela imuno-histoquímica. Portanto, nossos dados suportam a hipótese de que o córtex do camundongo SOD1(G93A) não recapitula a doença nos pacientes, e recomendamos o uso de um modelo diferente para estudos futuros da patologia cortical da ELA.
A degeneração característica dos neurônios motores (MNs) na ELA inicialmente causa atrofia muscular progressiva, levando a dificuldades de movimento, fala e deglutição e insuficiência respiratória no estágio final. Geralmente, a ELA é considerada uma doença multifatorial com mecanismos patológicos pouco compreendidos e com sobrevida média de três a cinco anos. Atualmente não há cura ou prevenção disponível, apenas tratamento sintomático.
Embora os MNs sejam reconhecidos como o principal tipo de célula afetada pela patologia, vários estudos confirmaram que também as células não neuronais, incluindo a glia, sofrem alterações e participam da progressão da ELA1,2. As células gliais respondem à neurodegeneração ou lesão por vários mecanismos. A micróglia e os astrócitos adquirem o chamado estado reativo marcado por mudanças na expressão gênica e na morfologia. Pela ativação de vias inflamatórias e anti-inflamatórias, eles protegem o tecido de danos adicionais. No entanto, na fase crônica das doenças, eles têm um efeito prejudicial e contribuem para a progressão da neurodegeneração. Tradicionalmente, os astrócitos reativos e a micróglia eram divididos em subtipos A1 e A2 ou M1 e M2, respectivamente. No entanto, essa terminologia é atualmente considerada desatualizada, pois várias subpopulações de células gliais com assinaturas de genes específicos foram recentemente descritas em diferentes modelos de doenças3,4,5. Astrócitos associados à doença (DAA), micróglia associada à doença (DAM), micróglia de resposta ativada (ARM) e micróglia de resposta ao interferon (IRM) são apenas alguns exemplos. Os oligodendrócitos, por outro lado, são mais suscetíveis a alterações patológicas. Em reação à doença, eles tendem a degenerar, em vez de se transformar em um estado reativo. No entanto, seu papel passivo na progressão da doença foi recentemente questionado por estudos que descrevem sua contribuição para a imunoproteção, sinalização do interferon e processamento e apresentação de antígenos6,7.
O papel dos astrócitos, microglia e oligodendrócitos, e suas respectivas alterações relacionadas à patologia, foram relatados em pacientes com ELA várias vezes8,9,10. A maioria dos dados disponíveis vem da medula espinhal, mas alguns estudos também relataram patologia glial no córtex11. As alterações patológicas conhecidas foram identificadas principalmente em tecido post mortem, o que não permite o estudo dos mecanismos da doença, aumentando a necessidade de um modelo animal confiável. Atualmente, o camundongo SOD1(G93A) representa o modelo mais amplamente utilizado semelhante a ALS12 familiar. Fenotipicamente, o modelo corresponde ao curso da doença, e estudos na medula espinhal e no tronco cerebral relataram alterações celulares relacionadas à ELA encontradas anteriormente em pacientes2,8,13,14,15,16. O córtex, no entanto, parece ser objeto de controvérsia. O número de estudos é limitado e os resultados sugerem desfechos contraditórios. Enquanto alguns mostram que células gliais corticais e MNs são afetados pelo fenótipo semelhante a ALS17,18,19,20, outros relatam nenhum efeito na área cortical no modelo SOD1(G93A)21.
